黄芪对黑曲霉植酸酶代谢活性的影响

摘要:利用FeSO4-钼蓝法研究了中草药黄芪(Astragalus membranaceus)粉末、黄芪水提取液对黑曲霉(Aspergillus niger)植酸酶在固体培养基和液体培养基中代谢活性的影响。结果表明,在0.03 g/mL的黄芪粉末和0.03 g/mL的黄芪水提取液作用下,44 h时黑曲霉中植酸酶活性最高。与对照相比,固体培养基中加入黄芪粉末和黄芪水提取液的吸光度分别提高了33.6%和37.8%,液体培养基中加入黄芪粉末和黄芪水提取液的吸光度分别提高了21.0%和28.7%。在最佳培养时间,植酸酶活性均随着黄芪浓度的增大而呈下降趋势,其中加入黄芪粉末的液体培养基变化幅度最大,植酸酶活性浓度0.01 g/mL是浓度0.05 g/mL的1.64倍;加入黄芪水提取液的液体培养基中植酸酶活性浓度0.01 g/mL是浓度0.05 g/mL的1.54倍;加入黄芪粉末和黄芪水提取液的固体培养基中植酸酶活性浓度0.01 g/mL分别是浓度0.05 g/mL的 1.50和1.42倍。

Effect of Astragalus membranaceus on Aspergillus niger Phytase Metabolic Activity

Abstract: The effect of traditional Chinese medicine Radix Astragali powder and water extract on Aspergillus niger phytase metabolic activity on solid and liquid culture medium were studied by using FeSO4-molybdenum blue method. The results showed that phytase activity was the highest if treated for 44 hours by 0.03 g/mL of astragalus powder and 0.03 g/mL of water extract. Compared with the control, the absorbance value was improved by 33.6% and 37.8% respectively by adding astragalus powder and astragalus extract water in solid culture medium; however, increase of 21.0% and 28.7% were respectively achieved in liquid culture. In the best culture period, phytase activity increased with the increasing of the concentration of astragalus. There was biggest change range in liquid medium which added astragalus powder.

水解植酸及其盐类的酶被称为植酸酶[1],它能使饲料中有机磷得到有效利用[2]。一般情况下为了满足畜禽对磷的需要,普遍采用的方法是在饲料中加入有机磷,但由此带来一些问题,例如消耗了大量的磷矿资源和若未被消化吸收的磷被动物排出体外,对土壤和水源造成污染。在饲料中加入植酸酶不但能提高动物生产性能,还能提高钙、镁、锌、锰、铜和铁的生物利用率[3]。植酸酶的研制和开发受到广泛重视。

黄芪(Astragalus membranaceus)是一种常用中药材[4],可促进黑曲霉(Aspergillus niger)中植酸酶的代谢。黑曲霉是一种常见的真菌,广泛分布于世界各地的粮食、植物性产品和土壤中[5],便于收集培养,而且黑曲霉是微生物中产植酸酶的高产真菌。试验研究了黄芪对植酸酶代谢的影响[6],得出了植酸酶活性的最佳培养时间和浓度,有利于提高植酸酶的利用率,提高经济效益。

1 材料与方法

1.1 供试材料

黄芪购自中药材市场,粉碎成粉末备用;10%黄芪水提取液的制备:准确称取固体黄芪10 g于250 mL烧杯中,加入50 mL去离子水,沸腾后煮20 min,冷却后用八层纱布过滤得到滤液,再次倒入烧杯50 mL去离子水,沸腾后再煮20 min,冷却后用八层纱布过滤的滤液和第一次的滤液混合后定容至100 mL,放置待用;黑曲霉由贵州大学农学院植物病理实验室提供;植酸酶活性测定采用FeSO4-钼蓝法进行[7]。

1.2 培养基的配制

1)不加黄芪的对照培养基。固体培养基的配制:麸皮20 g,硫酸铵4 g,去离子水定容至50 mL;液体培养基的配制:麸皮5 g,硫酸铵4 g,去离子水定容至50 mL。
