p38MAPK信号通路调控IL―6、HIF―1α及VEGF对慢性鼻―鼻窦炎发病机制的影响

[摘要]目的 观察p38丝裂原活化蛋白激酶(p38MAPK)在慢性鼻-鼻窦炎(CRS)不伴鼻息肉(CRSsNP)、CRS伴鼻息肉(CRSwNP)以及正常钩突黏膜组织中表达程度的差异以及不同类型的CRS中细胞因子白介素6(IL-6)、缺氧诱导因子-1α(HIF-1α)和血管绕ど长因子(VEGF)的表达,探讨并分析p38MAPK信号通路在CRS中可能的作用机制。方法 选取2015年7 月~2016年7月青岛市市立医院耳鼻喉科行鼻内镜手术患者,32例CRSsNP患者窦口组织、30例CRSwNP患者鼻息肉组织和24例正常钩突黏膜组织,分别采用蛋白质印迹法(Western Blot)检测p38MAPK在三组中的蛋白表达量、用逆转录聚合酶链式反应法(RT-PCR)检测IL-6、HIF-1α 和VEGF在三组中的信使核糖核酸(mRNA)的表达水平。结果 p38MAPK在CRSsNP、CRSwNP中的表达均高于正常黏膜组织,而两组实验组间的表达差异无统计学意义(P=0.143);IL-6在CRSsNP、CRSwNP中的表达均高于正常黏膜组织,而两组实验组间的表达差异无统计学意义(P=0.082);HIF-1α 和VEGF在CRSwNP中表达均明显高于正常组织(P

[Abstract]Objective To observe the difference of the degree of expression of p38 mitogen-activated protein kinase (p38 MAPK) in chronic rhinosinusitis (CRS) without nasal polyps (CRSsNP),CRS with nasal polyps (CRSwNP) and normal uncinate process,and the expression of interleukin-6 (IL-6),hypoxia-inducible factor 1 alpha (HIF-1α ) and vascular endothelial growth factors (VEGF) in different types of CRS.And explore the possible mechanism of p38MAPK signaling pathway in CRS.Methods Patients with nasal endoscopic surgery in department of otolaryngology of Qingdao Municipal Hospital were selected from July 2015 to July 2016 in Qingdao municipal hospital otolaryngology.32 cases of patients with CRSsNP sinus tissue,30 cases of CRSwNP patients with nasal polyps and 24 cases of with normal uncinate mucosa were selected for the study.The protein expression of p38 MAPK in three groups was detected by Western blotting.And the mRNA expression levels of IL-6, HIF-1α and VEGF in three groups were detected by reverse transcriptase-polymerase chain reaction (RT-PCR).Results The expression of p38MAPK in CRSsNP and CRSwNP was higher than that in normal mucosa, but there was no significant difference between the two groups (P=0.143).The expression of IL-6 in CRSsNP and CRSwNP was higher than that in normal mucosa,but there was no significant difference between two groups (P=0.082).The expression of HIF-1α and VEGF in CRSwNP was significantly higher than that in normal tissues (P
