Study on the effect of high intensity exercise and bamboo leaves extract on rats

Abstract. In order to enrich the theoretical study of high-intensity endurance training, explore new anti-fatigue supplements and broaden the applications of bamboo leaf extract, the effect of high-intensity endurance training and bamboo leaf extract on the blood biochemical parameters of rats was studied. 24 SD rats were randomly divided into quiet control group, sport control group and dosed sport group, with 8 rats in each. Sport control group experienced 6 week treadmill training. Dosed sport group experienced 6 week treadmill training and were fed 300mg / (kg ・ d) bamboo leaf extract solution. Quiet control group and sport control group were fed with the same volume of distilled water. After six weeks, the activity or content of rat’s serum alanine aminotransferase enzyme (ALT), aspartate amino transfer enzymes (AST), lactate dehydrogenase (LDH), creatine kinase (CK), blood glucose (Glu), hemoglobin (Hb), serumurea (BU), muscle glycogen and liver glycogen and exhaustive exercise time of sport control group and dosed sport group were measured. The results show exhaustive exercise causes significant decrease of Hb, liver glycogen, muscle glycogen and Glu, and significant increase of BUN, Bla, ALT, AST, LDH and CK, but taking bamboo leaf extract inhibited the downward trend of Hb, liver glycogen, muscle glycogen and Glu and upward trend of BUN, Bla, ALT, AST, LDH and CK and increase exhaustive exercise time by 28.87%.

High-intensity endurance training is a kind of physical training, focusing on long-term persistent exercise training, and it mainly aims to enhance the cardio-pulmonary function, and improve the muscle’s ability of taking advantage of oxygen, so that the body can adapt to long-term aerobic exercise, and it can also enhance the sports level of athletes. Endurance training supplement is a significant research direction of human movement science field, and various natural products including aloe, have been widely applied as the anti-fatigue product in endurance training [1-3]. However, there are few studies about the bamboo leaf extract. Bamboo is a kind of perennial evergreen with a wide coverage and huge output in China, which plays a significant role in people’s life and production, with profound cultural significance and practical value. The bamboo is full of valuables, for instance, bamboo stem can be used for making daily necessities, such as farm tools, furniture, etc. bamboo shoots can be eaten, and bamboo leaves can be used as fodder or fuel. In addition, in recent years, the physiological function, medicinal value and nourishing activity of bamboo leaves become a hotspot in the study on natural product field. [4] and the bamboo leaf extract has been gradually developed as the food additives, nutrition, cosmetics, drugs, etc. In order to enrich the theoretical study on the high-intensity endurance training, explore new anti-fatigue supplements and broaden the applications of bamboo leaf extract, the effect of high-intensity endurance training and bamboo leaf extract on the blood biochemical parameters of rats was studied.

1.2 The conflict of communicative language and method

And Westerners are more confident and like to show personality, when someone is on the praise, Westerners will choose to accept, and thank the applause. But Chinese are relatively modest, pleading in the face of others praise, or use some modest words to deal with.

1.3 The conflict of body language.

Body language is also a way of human communication, body language is more kind, compared to language exchange, express the meaning more widely. New research in psychology shows that, between the two body sizes can show the relationship of that, and some body movements can also determine the familiar extent of the two. If a couple of men and women hug, kiss, both have close relationship, but mostly exist lover relationship, they use body language to express their admiration. Due to cultural differences, cross-cultural communication conflicts exist in understanding body language, such as the Western friends meet, they need to hug or kiss to goodwill, but Chinese are used to expressing friendship with handshake, and Chinese think hug or kiss belongs to more intimate action, which will only between relatives, lovers; Americans think his tongue put out as a contempt for others, but Tibetan have different opinions, and show respect to the guest out their tongue. Because of the difference of body language understanding and the expression way, there is a certain amount of conflict sometimes in intercultural communication.

1. Material and method

1.1 Experimental raw materials and object

The bamboo leaf extract was purchased from Zhejiang University, and it is claybank powder, with more than 24% of general flavone glucoside. 30 healthy male Sprague-Dawley (SD) rates, about 180 to 220 g in weight were provided by Experimental Animal Center of Xi’an Jiaotong University College of Medicine. The ALT, AST, BU, Glu, Hb, liver glycogen, CK and LDH content or active detection kit were provided by Nanjing Jiancheng Bioengineering Institute. The main instrument was 721 visible spectrophotometer and ACE fully automatic biochemical analyser.

1.2 Experimental method

2.2.1 Experimental model

After the experimental animals are cultivated adaptively for 7 days, they will be selected by three stages with the 15m/min and 5min/d exercise amount, for eliminating some rats that cannot adapt to the treadmill training. The rest rats should be divided into three gourps randomly, eight for each group, including the quiet group, sports group and sports dosing group. The exhaustion judging standards [5] are: the animals could not keep up with the preset speed; the hips of rats were pressed on the back wall of the cage; the hind legs were dragged by the belt for 30 seconds; and it was ineffective to drive with hairbrush. The behavior characteristics were deep breath, large amplitude, tired expression, in prone position, and no response after stimulation.

Before training, adaptive exercise at 15 m/min was given for the rats for 5min, and then it was the official training. The training model was adjusted a little on the basis of the Bedford model [6]. The rats were trained for 6 w.

2.2.2 Medication

Dissolve the bamboo leaf extract in distilled water, about 300mg/ (kg・d) would be fed for the exercise dosing group, while distilled water would be given for the other two groups as contrast.

2.2.3 Preparation of animal samples and Index testing method

In the last day of the sixth week, the exhaustion time and weight of rats in sports group and sports dosing group were recorded, while the rats in quiet group were weighted. And then the rats were given moderate anesthesia with diethyl ether. The eyeballs were taken for collecting blood, which would be made into plasma or serum by adding corresponding anticoagulation according to the requirements of index. Meanwhile, whole blood was taken with capillary tube for testing blood lactic acid. After the blood was taken and placed in 37℃ water for 30min, it was centrifuged for 10 min in 2500-3000 rpm, and then the supernatant, namely the serum was extracted and reserved in 4℃ refrigerator.
