雄黄对MRL/lpr狼疮小鼠抗ds—DNA抗体和T细胞表面分子CD69、CD25表达的影响

【摘要】 目的:初步观察雄黄对MRL/lpr狼疮小鼠SLE动物模型抗双链DNA抗体及T细胞表面分子CD69、CD25表达及其生存时间的影响。方法:将MRL/lpr狼疮小鼠随机分为空白(蒸馏水)对照组、纳米雄黄低中高剂量组,每天灌胃一次,分别在不同时间点采集血清和称重;在实验结束后,取外周淋巴结分离T细胞并体外培养;放射免疫法测定血清抗ds-DNA抗体效价,流式细胞仪检测T细胞表面活化分子CD69、CD25表达水平;并计算各个观察点的小鼠死亡率。结果:雄黄治疗各组MRL/lpr小鼠生存时间比对照组明显延长,差异有统计学意义(P

【Abstract】 Objective:To investigate the effects of Realgar on anti ds-DNA antibody and the expression of CD69,CD25 on MRL/lpr mouse T cell.Method:MRL/lprmice were divided into four groups:control group,Low dose realgar group,Medium dose realgar group,and High dose realgar group separation the peripheral lymph node during treatment,all mice were respectively collected serum and weighing in each time.In the end,T cells were cultured in vitro,which were separated from the peripheral lymph node in MRL/lpr mouse.Anti dsDNA antibody in serum were detected by radioimmunoassay,flow cytometry were used to detect the express of CD69 and CD25 by activated T cells in vitro in response to Concanavalin(Con A).Result:The serum level of anti ds-DNA antibody in realgar groups were much lower than those in control group of MRL/lpr mice(P

雄黄为硫化物类矿物雄黄的矿石,又名黄金石。《神农本草经》记载:“黄金石,辛温,主寒热,鼠瘘,恶疮,疽痔,死肌,杀白虫毒。”《金匮要略》记载含有雄黄的升麻鳖甲汤用于治疗阴阳毒,而阴阳毒与系统性红斑狼疮(SLE)的症状很相似。中医临床上主要用于祛风除湿,解毒杀虫。雄黄既可入丸散作为复方使用,也可单独应用。近年来,雄黄及其复方应用于白血病治疗,取得肯定的临床疗效,其基础研究也获得了可喜的成绩,展示了广阔的应用前景,使人们重新认识雄黄。新近的研究表明,雄黄能增强内皮系统的吞噬功能,诱导肿瘤细胞凋亡[1-3];成静等[4]报道三氧化二砷对小鼠免疫功能有抑制作用;朱小春等[5-6]报道雄黄的主要成分之一三氧化二砷剂可以减轻MRL/lpr狼疮小鼠的临床症状,提示雄黄也可能具有治疗SLE的作用。然而国内外对雄黄的安全性和免疫调节作用机制研究很少。本实验初步观察了雄黄对SLE动物模型抗双链DNA抗体及T细胞表面活化分子CD69、CD25表达及其生存时间的影响,旨在探讨中药雄黄治疗狼疮的价值,并研究其具体的分子作用机制,现报告如下。
