家兔胆汁的光谱特性及降压效应观察

[摘要] 目的 观察家兔新鲜胆汁的光谱特性及其降压效应,为将其开发为临床降压药做前期研究。 方法 家兔新鲜胆汁稀释200倍后,直接在紫外光谱仪下测定;新鲜采集胆汁,干燥后压片,在红外光谱仪上测定其红外光谱;21只3月龄SD大鼠,随机分成3组:假手术组、模型组和胆汁组。模型组和胆汁组结扎大鼠左肾动脉建立肾性高血压模型,假手术组只分离左肾动脉而不结扎,其余操作一样。待大鼠基本康复后,模型组、假手术组给予生理盐水灌胃,胆汁组给予家兔新鲜胆汁灌胃,1个月后,用Med Lab生物信号采集系统测定大鼠血压值。 结果 紫外光谱中276、386 nm处各有一紫外吸收峰,强度分别是0.419和0.314;红外光谱在指纹区1 015.39~1 090.64 cm-1和918.26~968.40 cm-1处各有一组特征吸收峰。动物实验结果显示,模型组血压值比假手术组高,说明造模成功,而胆汁组血压值比模型组低且差异有统计学意义(P < 0.05),说明胆汁有降压作用,同时胆汁组大鼠体重较假手术组、模型组低。 结论 家兔胆汁有独特的紫外、红外光谱,可以作为鉴别家兔胆汁的可靠指标,同时家兔胆汁能有效降低肾性高血压大鼠的血压,具有开发为降压药的潜力。

Observation of spectral characteristic and antihypertensive effect of rabbit bile

[Abstract] Objective To observe the spectral characteristic and antihypertensive effect of rabbit bile and do some pilot research to exploit it into antihypertensive drugs. Methods Fresh rabbit bile ultraviolet (UV) spectrum was measured with ultraviolet spectrum apparatus after 200 times diluted. Infrared spectrum was measured with infrared spectrum apparatus after completely dried. 21 SD rats aged 3 months were divided into three groups randomly, which were model group, bile group and sham group. Model group and bile group rats were ligated left kidney artery under general anaesthesia, then sham group rats were conducted with the same surgery except for ligating kidney artery. After healing, saline was administrated to the model and sham group rats by po, meanwhile, fresh rabbit bile was administrated to bile group rats by po. One month later, blood pressure was measured with Med Lab biosignal collecting system. Result UV curve of rabbit bile had absorption peak at 276 nm and 386 nm, the strength was 0.419 and 0.314 respectively. Infrared curve had two groups characteristic absorption peaks during 1 015.39-1 090.64 cm-1 and 918.26-968.4 cm-1 in finger print district. Data of animal experiment displayed the mean arterial pressure of model group was higher than that of the sham group (P < 0.05), which indicated the hypertension model was built successfully. Compared with model group, the mean arterial pressure of bile group decreased significantly (P < 0.05), which indicated bile had effect on antihypertensive. Meanwhile the weight of bile group rats was lower than that of the model group and sham group. Conclusion Rabbit bile has special UV and infrared apectrum, which can be used to discriminate and identify the rabbit bile, and rabbit bile can decrease rats blood pressure, which suggests rabbit bile can be utilized as antihypertensive drugs.
