CD133、CD44和Notch1在卵巢上皮肿瘤中的表达研究

[摘要] 目的 探讨CD133、CD44和Notch1在卵巢癌中的表达及其临床病理学意义。 方法 采用免疫组织化学ElivisionTMplus两步法,研究46例卵巢癌、18例卵巢囊腺瘤和10例正常卵巢上皮组织中CD133、CD44和Notch1的表达情况。 结果 在正常卵巢上皮组织、卵巢囊腺瘤及卵巢癌中CD133、CD44和Notch1阳性表达率分别为10.0%、27.8%、60.9%,0、11.1%、47.8%及20.0%、38.9%、63.0%,三者在三组间表达差异均有统计学意义(均P < 0.05)。卵巢癌中CD133及CD44阳性表达与卵巢癌的临床分期、病理分级及淋巴结转移相关,Notch1阳性表达与卵巢癌的病理分级及淋巴结转移相关(P < 0.05)。相关性分析显示,在卵巢癌中CD133、CD44与Notch1,CD133与CD44间的表达均呈正相关(r = 0.401,P < 0.01;r = 0.307,P < 0.05;r = 0.589,P < 0.001)。 结论 CD133、CD44和Notch1在卵巢癌中的阳性表达率明显高于卵巢囊腺瘤与正常卵巢上皮组织,提示这三者的高表达可能与卵巢癌的发生、发展有一定的联系。

Study on the expression of CD133, CD44 and Notch1 in ovarian epithelial carcinoma

[Abstract] Objective To investigate the expression of CD133, CD44 and Notch1 in epithelial ovarian cancer and its clinical pathology meaning. Methods The expression of CD133, CD44 and Notch1 were detected in 46 cases of ovarian epithelial carcinoma, 18 cases of benign ovarian epithelial tumor and 10 cases of normal ovarian epithelial tissue by ElivisionTMplus two-step of immunohistochemical staining. Results The positive expression rates of CD133, CD44 and Notch1 in normal ovarian epithelial tissue, benign ovarian epithelial tumor and ovarian epithelial cell carcinoma were 10.0%, 27.8%, 60.9%; 0, 11.1%, 47.8% and 20.0%, 38.9%, 63.0% respectively, statistical differences among the three groups were significant (P < 0.05). The expression of CD133 and CD44 protein were related to clinical stage, pathological grade and lymph nodes metastasis; the expression of Notch1 protein was related to pathological grade and lymph nodes metastasis (P < 0.05). In addition, the expression of CD133 protein was positively related to that of CD44 and Notch1 protein (r = 0.589, P < 0.001; r = 0.401, P < 0.01). The expression of CD44 protein was positively related to that of Notch1protein (r = 0.307, P < 0.05). Conclusion The positive expression rates of CD133, CD44 and Notch1 in ovarian epithelial carcinoma are higher than those in the normal ovarian epithelial tissue and benign ovarian epithelial tumor, which prompts that the high expression of CD133, CD44 and Notch1 may involve in the carcinogenesis process of ovarian epithelial carcinoma.
